Food Texture Analysis

Texture testing is a well-established technique for evaluating the mechanical and physical properties of raw ingredients, food structure, and designs, and for pre- and post-quality control checks. Texture testing has applications across a wide range of food types, including baked goods, cereals, confectionaries, snacks, dairy, fruit, vegetables, gelatins, meat, poultry, fish, pasta, and even pet food. Since texture is a property related to the sense of touch, it can be measured easily by mechanical methods in units such as force. In texture testing, standard tests such as compression, tension, and flexure are used to measure hardness, crispiness, crunchiness, softness, springiness, tackiness, and other properties of food. Comparing the results from mechanical texture analysis with trained human sensory panels has shown that the measurements have a high correlation with the various sensory attributes associated with textural quality…

Update on ETD and ECD for Proteomics

Electron capture dissociation (ECD) and electron transfer dissociation (ETD) are new tools that the mass spectrometry community is learning to apply to life science research. Their principal advantage is that the addition of an electron to the analyte causes very rapid ssion of a chemical bond. When applied to peptides and proteins, this leads to fragmentation that is largely randomized with respect to peptide sequence. In contrast, collisionally activated dissociation (CAD), where a target ion collides with a gas molecule, takes place over a 1000× longer time period. During this time, the energy of collision can be transferred through the molecule, leading to cleavage of the weakest bonds, which are often those attaching post-translational modifications (PTMs) such as carbohydrates and phosphates. While CAD does provide information-rich mass spectra, it may not contain information about sites of some modi cations, since signals from some structural elements may be missing…

Reproducibility of a Fractionation Procedure for Fish Muscle Proteomics

One of the challenges of comparative proteomics is the ability to separate and quantify all the components of highly complex mixtures of proteins, despite the fact that the concentration of their components can span several orders of magnitude. Subcellular fractionation procedures for muscle tissue have been used for some time in both gel and MS-based proteomics to address this situation, simplifying analysis by reducing the number of proteins in a given extract and improving the dynamic range by allowing larger loads per protein. Additionally, interpreting the observed changes in protein abundance may be eased by isolating a subpopulation of the whole tissue proteome. Specific examples of the use of these techniques for proteomic analysis include organelle and nucleus isolation, enrichment of extracts for secretory or membranar proteins, and albumin/immunoglobulin depletion of serum samples. On the other hand, since its use implies a greater number of sample processing steps than doing the whole extract, it is expected that some noise may be introduced by a fractionation procedure. Therefore, it seems important for comparative proteomic experiments to simultaneously ensure that no interbatch bias is introduced and that intrabatch noise is minimized, so that the benefits of performing this step outweigh its disadvantages…
