| Reproducibility of a Fractionation Procedure for Fish Muscle Proteomics |

One of the challenges of comparative proteomics is the ability to separate and quantify all the components of highly complex mixtures of proteins, despite the fact that the concentration of their components can span several orders of magnitude. Subcellular fractionation procedures for muscle tissue have been used for some time in both gel and MS-based proteomics to address this situation, simplifying analysis by reducing the number of proteins in a given extract and improving the dynamic range by allowing larger loads per protein. Additionally, interpreting the observed changes in protein abundance may be eased by isolating a subpopulation of the whole tissue proteome. Specific examples of the use of these techniques for proteomic analysis include organelle and nucleus isolation, enrichment of extracts for secretory or membranar proteins, and albumin/immunoglobulin depletion of serum samples…

| Highlights From HCA 2010: Human Stem Cells Replace Animal Testing |

The Seventh Annual High Content Analysis (HCA) meeting attracted about 325 scientists to the historic Fairmont Hotel in San Francisco, CA, from January 12 to 15, 2010. The program consisted of three parallel lecture tracks plus about 35 posters. Breakout sessions provided opportunities for focused networking on particular topics, including Novel Probes and Sensors, HCA of Stem Cells, Data Management, and Toxicology Profiling. The HCA meeting typically attracts the leaders from core facilities scattered throughout the world. Thus, several of the major vendors organized users’ group meetings as part of the program. These are especially useful in networking. In particular, the early majority is able to seek out mentors from the early adopters, and early adopters can meet with the vendors’ technical staff to resolve problems and search for solutions to unmet needs…

NEW APPLICATION NOTE (AN23) details the ideal separation provided by TSK-GEL Amide-80 columns for the analysis of glycans.
Researchers at the University of Erlangen-Nuremberg have studied the role of TGFR-3 ZP glycosylation in order to achieve a better understanding of transduction pathways, especially for fibrosis or neurodegenerative diseases. They discovered that TSK-GEL Amide-80 column chemistry is ideally suited for the separation of charged and neutral fractions of glycan pools in a single run. The retention of fluorescence labeled (2-aminobenzamide, 2-AB) polysaccharides by a TSKgel Amide-80 column enables the identification of glycan structures by comparison to a labeled dextran ladder.
