In the era of high-throughput cell biology, experimentation occurs predominantly by executing cell-based assays within microplates. Assays range from routine, such as optimizing transfection conditions and culture conditions, to visualization and analysis of large multicellular structures that better mimic in vivo conditions. The growing number of innovative cellular assays challenges conventional microplate-based cellular analysis systems. There is an increasing requirement to leverage brightfield and multicolor fluorescence capabilities to perform high-throughput morphologic and functional analysis of both living and fixed cells, without the time, cost, and complexity associated with high-content systems. Key constraints include 1) limited utility of transmitted light imaging modes in microplates at the walls of the wells; 2) inability to scan large areas of microplate wells in <30–40 min; and 3) requirement for a dedicated, experienced operator for effective use of complex cellular analysis systems for routine assays…
